Kornfield is van joodse afkomst en heeft vier broers. Hij is een twee-eiige tweeling. Zijn vader was een wetenschapper, waardoor hij geïnteresseerd raakte in genezing, geneeskunde en wetenschap. Hij volgde een cursus Aziatische filosofie bij Dr. Wing-tsit Chan. Kornfield studeerde uiteindelijk boeddhistische Aziatische studies. Na zijn afstuderen aan het Dartmouth College in 1967, trad Kornfield toe tot het Peace Corps en werd hij naar Thailand gestuurd, waar hij werkte in teams voor tropische geneeskunde in de Mekong River-vallei. Daar ontmoette hij en werd een monnik onder de bosmeester Ajahn Chah, en oefende later met Mahasi Sayadaw van Birma en Dipa Ma. Kornfield keerde in 1972 terug naar de Verenigde Staten en nam in de zomer van 1974 deel aan de oprichtingssessie van de Naropa University. Uit de verenigingen van deze periode kwam de Insight Meditation Society, mede opgericht in 1975 met Sharon Salzberg enJoseph Goldstein in Barre, Massachusetts. In 1987 was hij mede-oprichter van Spirit Rock Meditation Center in Woodacre, Californië. Kornfield heeft veel van de Vipassana-leraren in Amerika opgeleid en samen met de Dalai Lama en wereldwijd bijeenkomsten georganiseerd en geleid voor boeddhistische leraren. Opgeleid met een Ph.D. in de klinische psychologie van Saybrook Institute, Kornfield heeft uitgebreid geschreven over de brug tussen oosterse en westerse psychologie. Zijn dochter Caroline is afgestudeerd aan Berkeley Law en beoefent asielrecht. Zijn ex-vrouw Liana is kunstenaar en therapeut. Zijn vrouw Trudy Goodman  is ook een gerenommeerd meditatieleraar en de grondlegger van InsightLA, dat training in Vipassana combineert met niet-sektarische mindfulness- en compassiepraktijken, waaronder Mindfulness-Based Stress Reduction (MBSR) en Mindful Self- Compassie (MSC).
